In Vitro Cytotoxicity of Some Thai Medicinal Plants and Daphnane Diterpenoid from Trigonostemon redioides

Screening for cytotoxicity using the crystal violet colorimetric assay of 36 extracts from 10 Thai plants belonging to 9 families resulted in the selection of an herb extract:the dichloromethane extract of the roots of Trigonostemon redioides (Kurz) Craib, which exhibited cytotoxicity against HuCCA-1 and KB cells. The extract was selected for further detailed investigation to isolate and characterize the bioactive constituents by bioassay-guided fractionation of cytotoxicity against HuCCA-1 and KB cell lines viability as a model. The roots of T. redioides were purchased in Loei Province. Sequential extraction of each plant material was carried out with hexane, dichloromethane and ethanol. The dichloromethane extracts were evaluated by cytotoxicity testing and purified by chromatographic separation. The isolated compounds were identified by physical and spectroscopic methods, resulting in a daphnane, rediocide A (1), (+)-syringaresinol, scopoletin,tomentin, and stigmasterol.
